摘要
目的了解普伐他汀对自发2型糖尿病大鼠胰岛素抵抗、血浆NO浓度和视网膜组织超微结构的影响,探讨普伐他汀对糖尿病视网膜病变的预防作用。方法 6周龄大鼠随机分为:自发糖尿病组,正常对照组,普伐他汀治疗组3组,每组18只。前两组正常饮食,普伐他汀治疗组在正常饮食中加入普伐他汀(100mg·kg-·1d-1)。分别于10、20、30周测量各组鼠的体质量、摄食量,并检测空腹血糖、胰岛素和血浆中NOX及瘦素的浓度;于30周龄处死各组大鼠,采用HE染色和透射电镜观察视网膜超微结构改变。结果与正常对照组相比,自发糖尿病组大鼠各时段体质量、摄食量明显增加;瘦素水平和IR值明显增高;20和30周龄大鼠血浆NOX平均浓度明显降低;视网膜变薄,神经节细胞数目减少;毛细血管基底膜明显增厚,视杆细胞的膜盘排列紊乱,间隙增宽。而普伐他汀治疗组各时段体质量、摄食量被明显控制;血浆NOX浓度无明显下降;20和30周龄大鼠瘦素水平和IR值的提升被显著削弱;视网膜超微结构的改变都不明显。结论普伐他汀能够在一定程度上控制自发糖尿病大鼠体质量和摄食量的增加;能够降低血中瘦素水平;提高血浆中NOX浓度,阻止糖尿病的发展。同时,普伐他汀能够控制和延缓糖尿病视网膜病变的发生。
Objective To investigate the impacts of pravastatin on progressive insulin resistance (IR),plasma level of nitric oxide(NO) and retinal ultramicrostructure in a rat model of Type Ⅱ Diabetes,and to investigate the preventive effect of pravastatin in the pathological changes in diabetic retina.Methods The 6-week-old diabetes rats were randomly divided into three groups with 18 in each:the untreated gourp,the normal control group and the pravastatin-treated group.The rats in the first two groups were fed with normal diet and the rats in the pravastatin-treated group were fed with normal diet plus pravastatin (100 mg·kg-·1d-1).The food intake and body weight,as well as the plasma levels of glucose,insulin,NOX leptin were measured in 10,20 and 30 weeks of age,respectively.In 30 weeks of age,rats in all the three groups were sacrificed.The paraffin slices and ultra-thin slices of retina were prepared and observed under light microscope and transmission electron microscope.Results In the untreated diabetes rats,the body weight and food intake increased obviously when compared with the normal control,the leptin levels and IR values were significantly increased.In 20 and 30 weeks-old diabetes rats,the average plasma NOX levels were obviously decreased,the retina became thin,the number of ganglion cells reduced,the capillary basement membrane thickened,the rod cell membrane disc arranged irregularly and the gap widened.In the rats treated with pravastatin,the body weight and food intake reduced obviously,the plasma NOX levels decreased not significantly,the decrease of leptin levels and IR value were significantly reduced in 20 and 30-week-old rats.There was no significant change in retinal ultrastructure of pravastatin treated rats.Conclusion Pravastatin could control the increase of body weight and food intake of spontaneously diabetic rats,reduce plasma leptin levels,increase plasma NOX concentration and prevent the development of diabetes.Pravastatin could control and delay the occurrence of diabetic
