摘要
[目的]探讨在屏障环境实验动物设施运行过程中应用真空高压灭菌等方法对于控制微生物污染的效果,以便达到动物实验环境控制标准。[方法]2009年,在山东省疾病预防控制中心实验动物中心屏障环境设施,对室内空气、饲料、垫料、饮水、工作服、饲养盒分别采用高锰酸钾和甲醛熏蒸、真空高压、消毒液浸泡进行灭菌,并观察其效果。[结果]采用双扉真空灭菌器对饲料132℃灭菌7 min、垫料132℃灭菌6 min、工作服132℃灭菌6 min,饮水采用液体121℃灭菌30 min,饲养盒121℃高压灭菌30 min或消毒液浸泡过夜,在使用周期范围内无菌保存均无菌生长。[结论]在屏障环境中对实验动物设施采用真空高压灭菌等方法进行灭菌对于控制微生物污染有良好的效果。
[Objective]To study the effect of the vacuum high pressure sterilization on microbial control for the laboratory animal facility in barrier environment during the operation,so as to reach the standard of animal experiment environment.[Methods]In 2009,the experiment was processed at the Animal Experiment Center,Shandong Center for Disease Control and Prevention.fumigation,autoclaving or disinfection and microbiological monitoring was conducted on room air,feeder,bedding,drinking water,clothing,cages,etc.to observe the effect at different time.[Results]The double-gate vacuum sterilizer was used for sterilization on feeder 7min 132℃,bedding sterilized 6min 132 ℃,gown sterilization 6min 132 ℃,drinking water liquid pressure sterilization 30 min 121 ℃,sterile effect was achieved,the cages in the sterile storage cycle within the life cycle with disinfectant,sterilization effect could be achieved overnight.[Conclusion]In barrier enviorment,the microorganism pollution can be well controlled by the vacuum high pressure sterilization method.
