摘要
以杜仲幼嫩叶片为试材,在MS为基本培养基上添加不同浓度的生长素和细胞分裂素,对愈伤组织诱导的优化条件进行了研究。结果表明:2,4-D和6-BA对诱导杜仲叶片愈伤组织诱导均有影响,最优培养基为MS+2,4-D 0.5 mg/L+6-BA 0.3 mg/L,诱导率达87.5%,且愈伤组织生长较好。培养基中添加5 g/L活性炭能有效地抑制外植体褐化。
To study the optimum conditions of callus induction by using the leaves of Eucommia ulmoides Olive as explants,and MS being the basic medium supplemented with different concentrations of auxin and cytokinin.The results showed that both 2,4-D and 6-BA had impact on callus induction.The optimum medium type was MS+2,4-D 0.5 mg/L+6-BA 0.3 mg/L,which could lead a high inducing rate of 87.5% and was good for the growth of callus.5 g/L activated charcoal(AC)could effectively inhabit the explants browning.
