高光学纯度INCB018424的合成

摘要 对Janus激酶抑制剂INCB018424的重要中间体(3R)-3-(4-溴-1H-吡唑-1-基)-3-环戊烷丙氰(R-8)的纯化方法进行了改进,通过重结晶[-10℃下,以V(乙醚)∶V(石油醚)=1∶20为重结晶溶剂]的方法使R-8的ee值由84.0%提高至97.0%。由纯化的R-8合成了ee值达99.9%的INCB018424。其结构经1H NMR表征。 Purification method of important intermediate of Janus kinase inhibitor INCB018424,(3R)-3-(4-bro-1H-pyrazol-1-yl)-3-cyclopentylbutanenitrile(R-8),was improved and the ee value of R-8 can easily reach 97.0% from 84.0% by recrystallization[at-10 ℃,V(ether) ∶ V(petroleum ether)=1 ∶ 20 as a recrystallization solvent].INCB018424 with 99.9%ee was synthesized from purity R-8.The structure were characterized by 1H NMR.
