摘要
目的了解慢性乙型肝炎(CHB)患者外周血单个核细胞(PBMCs)中T细胞受体重排删除环(TRECs)含量和PBMCs中初始T细胞比例,分析两者的相关关系。方法采集CHB患者和健康人外周静脉血标本,(1)应用实时荧光定量聚合酶链反应(RT-PCR)技术检测TRECs含量;(2)利用荧光标记抗体FITC-CD3、PE-CD45RA、PE-cy5-CD62L标记细胞表面分子,流式细胞仪检测PBMCs中CD3+CD45RA+CD62L+细胞比例;(3)TRECs含量与初始T细胞相关性分析。结果 (1)CHB组TRECs含量为(6.78±1.06)拷贝/103 PBMCs,低于对照组(8.16±2.07)拷贝/103 PBMCs,P<0.01。(2)CHB组初始T细胞比例为(20.73±6.75)%,低于对照组(27.82±9.66)%,P<0.05。(3)CHB组初始T细胞比例与TRECs含量呈正直线相关,相关系数r=0.688,P<0.01。对照组初始T细胞比例与TRECs含量正相关,r=0.646,P<0.01。两组r值比较差异无统计学意义,P>0.05。结论 (1)CHB患者PBMCs中TRECs含量与初始T细胞比例均低于正常人;(2)CHB患者与正常人PBMCs中TRECs含量与初始T细胞比例均正相关。
Objective To test the content of T cell receptor excision circles(TRECs) and the frequency of naive T lymphocytes in patients with chronic HBV infection and analyze their correlation.Methods Peripheral blood were collected from normal individuals and patients with chronic HBV infections.(1) TRECs were assessed by real-time PCR;(2) Lymphocytes were labeled with fluorescent-labeled antibodies FITC-CD3,PE-CD45RA and PE-cy5-CD62L and flow cytometry was used to detect the expressions of CD3,CD45RA and CD62L on the surface of T lymphocytes.Results(1) The TRECs(6.78±1.06) copies/103 in patients(n=25) with chronic HBV infections was significantly lower than that(8.16±2.07) copies/103 in healthy controls(n=25)(P0.01).The frequency of nave T cells(20.73±6.75)% in patients(n=25) with chronic HBV infections was significantly lower than that(27.82±9.66)% in healthy controls(n=25)(P0.05).(2) Both in patients with chronic HBV infections and in healthy controls,the frequency of nave T cells were in positive linear correlation with the TRECs,while the coefficient correlation were 0.688(P0.01) and 0.646(P0.01) respectively.The coefficient correlation in the two groups had no statistical differences,(P0.05).Conclusion Both the nave T cell in peripheral blood and the TRECs in patients with chronic HBV infection were decreased compare to the healthy adult,and the two indices had better linear correlation.
