摘要
以L-天冬氨酸为单体进行缩聚反应所得到的高分子量聚琥珀酰亚胺(PSI)为母体,己二胺为交联剂,采用乙醇胺对其进行改性,制备了羟化聚天冬氨酸水凝胶。研究了反应温度、交联剂及乙醇胺用量对水凝胶溶胀性能的影响;以水杨酸为模型药物,研究了其载药性能及在不同释放介质中的释放行为。结果表明,反应温度为40℃,交联剂用量为PSI用量的7%时水凝胶的溶胀性能最好。随着乙醇胺用量的增加,水凝胶在乙醇溶液中的溶胀倍率增加,当乙醇胺与PSI的物质的量比为0.5∶1时,溶胀倍率可达到145倍,载药量由最初的4.23 mg提高到29.92 mg。
The polyaspartic acid hydrogel with hydroxyl was synthesized using the high molecular weight polysuccinimide(PSI) as the matrix,hexamethylene diamine as crosslinking agent,and modified by ethanolamide.The PSI was synthesized by polycondensation reaction with L-aspartic acid as the monomer.Effects of reaction temperature,the amount of cross-linking agent and ethanolamine on the swelling behavior of hydrogel were studied.Salicylic acid was chosen as drug-model,its releasing behavior in different medium was studied.The results show that the swelling ratio of hydrogel gets the max value when the reaction temperature is 40 ℃, cross-linker dosage is 7% of PSI.With the increase of ethanolamine,the swelling ratio of hydrogel in ethanol solution increases,it can reach up to 145 times when the molar ratio of ethanolamide to PSI is 0.5∶1,and the drug loading amount increases from the initial 4.23 mg to 29.92 mg.
