摘要
以甲壳素为原料制成脱乙酰度为60.9%、相对分子量为7.96×105的壳聚糖,再用该壳聚糖与五氧化二磷、甲磺酸反应合成了1#至7#壳聚糖磷酸酯,用电位滴定法测定取代度,并用FT-IR进行分子结构表征;进一步将壳聚糖磷酸酯与碳酸钾作用制备1#至7#壳聚糖磷酯钾,并将壳聚糖磷酸酯钾配制成0.5、1、2、5、10 mg/L溶液,以蒸馏水为对照,进行台湾四九菜心、水稻种子的浸种发芽实验.浸种时间24~39 h,浸种温度24~30.5℃,每只培养皿50粒,每组重复3次.生物活性实验结果表明:1#、3#、4#壳聚糖磷酸酯钾对菜心、水稻种子的发芽没有促进作用,而7#壳聚糖磷酸酯钾对受体种子发芽的促进作用不明显;在质量浓度为0.5~10 mg/L时,2#、5#、6#壳聚糖磷酸酯钾对菜心种子的发芽率分别比对照组增长3%、16%、15%,对水稻种子的发芽率分别比对照组增长10%、10%、9%.
Chitin as raw material, Chitosan wich characterized with a degree of deacetylation of 60.9% and a molecular weight of 7.96 × 10^5 was obtained . 1^# -7^#Chitosan Phosphate were synthesized with Chitosan, P2O5 and MeSO3H. Their degree of substitution were determined by potentiometric titration and structures were characterized by FT-IR. 1^# - 7^# Chitosan phosphate were further reacted with K2 CO3 to produce 1^# - 7^# potassium Chitosan phosphate. The Potassium Chitosan Phosphate were prepared into solutions of 0, 5,2, 5,10 mg/ L. Taiwan four-nine sweet cabbage and Oryza sativa were applied in the trial, with thrice repeats for each group, 50 seeds in each petri dish, soaking time from 24 hours to 39 hours from species to species , the distilled water as control, The culture temperature was 24 - 30.5 ℃, 1^#、3^#、4^# Potassium Chitosan phosphate had no promotive effect to Taiwan four-nine sweet cabbage and Oryza sativa. 7^# had no significant as compared with the control . The concentration ranged from 0.5 mg/L to 10 mg/L ,2^#、5^#、6^#Potassium Chitosan phosphate increased the germination rate of the seeds of Taiwan four-nine sweet cabbage by 3% ,16% ,15% and increased the germination rate of the seeds of Oryza sativa by 10% ,10% .9% as compared with the control.
