摘要
目的 观察丹参对慢性低氧性肺动脉高压大鼠肺组织碱性成纤维细胞生长因子(bFGF)表达的影响,探讨丹参阻抑肺腺泡内动脉(IAPA)重建的机制。方法 Wistar大鼠随机分为3组:正常对照组、慢性低氧组、低氧丹参治疗组。低氧大鼠置于常压低氧舱3周后,测定血流动力学;观察IAPA结构变化,用免疫组化检测肺组织bFGF表达;用氯胺T比色法测定肺组织羟田氨酸含量。结果 丹参能降低平均肺动脉压,减少肺组织羟脯氨酸含量(P<0.05,P<0.01),抑制IAPA平滑肌细胞增生,降低肺组织bFGF表达强度 结论(1)丹参能抑制IAPA平滑肌细胞增生及胶原沉积而阻抑IAPA的结构重建。(2)抑制bFGF表达是丹参调节IAPA平滑肌细胞增生的重要途径之一。
Objective To evaluate the effect of SM on expression of bFGF in rat lung tissue of chronic hypoxic pulmonary hypertension. Methods 30 Rats were divided into 3 groups;hypoxic group, SM treatment group and control. On the 21 th day of hypoxia,the hemodynamics were measured and the intra-acinous pulmonary arteries (IAPA) changes were studied under light microscopes; the contents of hy-droxyproline(Hyp) in lung tissue were determined with calorimetric method after oxidized by chloramine T; immunohis-
tochemical staining with monoclonal anti-boby against human recombinant bFGF was perfomed in the paraffin section of rat lung. Results SM could reduce pulmonary artery mean pressure (mPAP) and the contents of Hyp in rat lung tissue (P< 0. 05, P < 0. 01 respectively ) , decrase expression of bFGF in wall of IAPA and inhibit hyperplasia of smooth muscle and collagen deposition. Conclusion SM could decrease expression of bFGF in wall of IAPA to inhibit remodelling of IAPA.
