磷脂型DHA对肿瘤细胞凋亡的影响 被引量:7

摘要 目的探讨磷脂型DHA(DHA-PC)的抗肿瘤生长作用。方法应用超临界CO2萃取脱除胆固醇技术从鸢乌贼卵中提取DHA-PC。采用MTT法、AO/EB荧光染色法和流式细胞术研究DHA-PC对人肝癌HepG-2细胞增殖、凋亡的影响,并进一步利用小鼠S180移植性肿瘤模型,验证DHA-PC的体内抑瘤效应。结果 DHA-PC能够抑制HepG-2细胞的增殖(P<0.05,P<0.01),诱导细胞凋亡,干扰细胞周期,使细胞停滞在G0/G1期;明显抑制荷瘤小鼠肿瘤的生长,其平均抑瘤率达40.94%。结论 DHA-PC在体内和体外都对肿瘤的生长具有抑制作用。 Aim To study the role of docosahexaenoic acid-phosphatidylcholine in inhibiting tumor growth.Methods DHA-PC separeted from cuttlefish spawn by supercritical carbon dioxide was used to prepare unilayer liposomes.The in vitro anti-cancer effect of DHA-PC was evaluated in human hepatocellular carcinoma HepG-2 cell lines by MTT method.Cell cycle and morphological features of HepG-2 cell were observed by flow cytometery and AO/EB staining,respectively.Moreover,The effects of DHA-PC on anti-tumor in vivo were investigated in transplanted tumor-bearing mice.Results DHA-PC could effectively inhibit the proliferation of HepG-2 cell(P0.05,P0.01),induce apoptosis of tumor cells and block the HepG-2 cell at G0/G1 phase;In vivo DHA-PC could inhibit the growth of tumor in mice,with an average inhibitory rate of 40.94%.Conclusion ZDHA-PC has strong anti-tumor activity both in vitro and in vivo and is a potential source of natural anti-tumor products.
