摘要
本文介绍了提高ＰＣＲ产物及其克隆效率的方法与经验：如，引物的设计、目的片段的回收、连接体系、利用引物上所设计的酶切位点克隆、通过平末端克隆，以及加快实验进程的技巧、出现问题如何设计查找原因等。
In order to improve the PCR products and its further cloning efficiency, we introduce several methologies on primer design, recovery of target DNA fragment, ligation system, etc. Cloning through the restriction enzyme incorporated in the primer, blunt-end ligation, several knacks in our work are also presented in this paper.
