摘要
以ACQUITY UPLC BEHAmide柱为分析柱,乙腈(0.2%TEA)和水作为流动相,采用蒸发光散射检测器,建立了超高效液相色谱分析烟草中鼠李糖、木糖、果糖、甘露糖、葡萄糖、蔗糖、麦芽糖的方法。7种水溶糖的加标回收率在85.2%~105.3%之间,相对标准偏差皆小于5.7%。方法可用于烟草及其制品中水溶性糖含量的测定。采用此法测定了30个烟草样品,以7种水溶性糖为指标对样品进行了聚类分析。结果表明通过烟草水溶性糖的聚类分析可以区分不同品种的烟样。
A new method for the determination of seven carbohydrates(rhamnose,xylose,fructose,mannose,glucose,sucrose,maltose) in tobacco by Ultra Performance Liquid Chromatography with an evaporative light scattering detector is studied.The carbohydrates were separated on an Acquity UPLC BEH Amide column by using acetonitrile-water(0.2%TEA) as the mobile phase.Recoveries of the method for the seven carbohydrates were from 85.2 % to 105.3 % with relative standard deviations(RSD)(n=6) of 0.78%~5.7 %.This method can be applied to the determination of carbohydrates in tobacco with good results.The contents of seven carbohydrates in 30 tobacco samples were determined,and the data were processed with cluster analysis.The results showed that the cluster analysis could be used in the comparison and classification of different kinds of tobacco.
