摘要
根据杜氏利什曼原虫(L.donovani)与婴儿利什曼原虫(L.infantum)基因组核苷酸序列之间具有高度的同源性的特点,设计合成了酸性核糖体蛋白-1(ARP-1)基因序列特异性的引物,应用多聚酶链反应(PCR)技术,以杜氏利什曼原虫的基因组DNA为模板,扩增获得了杜氏利什曼原虫的ARP-1基因克隆。序列分析表明,杜氏利什曼原虫与婴儿型利什曼原虫ARP-1基因的核苷酸序列之间的同源性为93%,编码的氨基酸残基序列的同源性为98.0%。
According to the high homolgy of DNA sequence between Leishmania donovani and Leishmania infantum,specific primers for acidic ribosomal protein1 (ARP1) DNA were designed.Using genomic DNA from Leishmania donovani as the template,ARP1 DNA fragment was amplified by polymerase chain reaction (PCR) and subcloned into pCR 21 (Invitrogen) and sequenced,Sequence analysis of ARP1 from Leishmania donovani and Leishmania infantum indicated 93% homology at nucleic acid sequence level,and 98% similarity at amino acid sequence level.
