摘要
建立了毛细管电泳法拆分2-羟基-3-甲氧基-3,3-二苯基丙酸的方法。考察了背景电解质的pH值和浓度、手性选择剂的种类及浓度、有机改性剂种类及浓度对分离的影响,并对分离条件进行了优化。实验结果表明,在60 mmol/L磷酸氢二钠(pH=9.10)为运行缓冲溶液,含35.7 mmol/L羟丙基-β-环糊精和5%甲醇的体系中,对映体达到基线分离。S型及R型对映异构体均在5~160μg/mL浓度范围内线性关系良好,平均加标回收率为100.3%,检出限为1.01μg/mL。
The enantiomers of 2-hydroxy-3-methoxy-3,3-diphenylpropionic acid were successfully separated by high performance capillary electrophoresis using HP-β-CD as a chiral selector.Effects of the pH and concentration of buffer,concentration of HP-β-CD,type and concentration of organic modifier on the resolution of enantiomers were investigated.Under the optimized conditions pH=9.10 60 mmol/L of phosphate buffer solution,35.7 mmol/L HP-β-CD and 5% CH3OH,the enantiomers of 2-hydroxy-3-methoxy-3,3-diphenylpropionic acid were separated baseline.Calibration curve was linear over the range of 5~160 μg/mL,and the limit of detection was 1.01 μg/mL for target analyte.
