摘要
目的:探讨蛇六谷乙酸乙酯萃取物A1组分对大鼠胶质瘤C6细胞增殖的抑制作用及其作用机制。方法:用MTT比色法检测不同浓度A1组分对C6细胞增殖的抑制作用,RT-PCR法检测A1组分对C6细胞C-myc、bc l-2,p53mRNA基因表达的影响。结果:A1组分对C6细胞的增殖具有明显的抑制作用,且呈量-效关系,并有明显的细胞形态学改变;RT-PCR结果显示,随着A1组分作用剂量的增加,C6细胞的C-myc、bc l-2及p53mRNA表达均有明显降低。结论:蛇六谷乙酸乙酯萃取物A1组分对大鼠胶质瘤C6细胞的增殖具有明显的抑制作用,其作用机制可能与抑制原癌基因C-myc、bc l-2及p53的表达有关。
Objective: To observe the inhibition of A1 component of konjac ethyl acetate extract on multiplication of rats' gliocytoma C6 cells and explore the mechanism.Methods: The inhibition of A1component of konjac ethyl acetate extract on multiplication of rats' gliocytoma C6 cells was determined by MTT method,the effect of A1component of konjac ethyl acetate extract on C-myc,bcl-2 and p53 expression in rats' gliocytoma C6 cells were tested by RT-PCR.Results: A1 component of konjac ethyl acetate extract could obviously inhibit the multiplication of rats' gliocytoma C6 cells,meanwhile showed significant dose-effect relationship,manifested obvious cytomorphology changes.The results of RT-PCR showed that the expression of C-myc,bcl-2 and p53 in rats' gliocytoma C6 cells was significantly reduced,that was gradually obvious with the increasing of dase.Conclusion: A1 component of konjac ethyl acetate extract can significant inhibit the multiplication of rats' gliocytoma C6 cells,which is related to down-regulating the expression of proto-oncogene C-myc,bcl-2 and p53 on gliocytoma C6 cells
