摘要
目的 :研究血管紧张素 转换酶 (ACE)基因多态性与血清 ACE水平及高血压的关系。方法 :采用多聚酶链反应对 2 16例观察对象 (正常人 10 3例 ,高血压患者 113例 )的 ACE基因进行分型 ,并测定血清 ACE活性。结果 :2 16例受检者中 ACE基因多态性与血清 ACE水平有关 ,ACE水平依次为 :DD型 >ID型 > 型 ,三者相互之间有显著性差异 ;多元逐步回归分析显示 ,ACE基因型决定不同个体血清 ACE活性的 2 7.5%的变化范围。结论 :当地人群 ACE基因多态性与血清ACE水平有关 。
Objective:To investigate the contribution of ACE genotype and their relation to the levels of serum ACE and hypertension in the local people.Method:Two hundred and sixteen subjects (103 healthy subjects and 113 hypertensives) underwent determination of the angiotensin coverting enzyme (ACE) genotype from leukocyte DNA by polymerase chain reaction and serum ACE levels.Result:A significant difference in serum ACE levels was observed among three ACE genotype classes.The highest level of serum ACE activity was in deletion homozygotes (genotype DD),and the lowest level in insertion homozygotes (genotype Ⅱ),and intermediate in heterozygotes (genotype ID).There was no difference in blood pressure among three genotypes both in hypertensives and normotensives,no difference of ACE alleles frequencies between the healthy and hypertensives.Conclusion:(1) ACE I/D polymorphism was associated with marked differences of ACE levels and accounted for 27.5 % of the variability of serum ACE levels.(2) No relationship was found between ACE I/D polymorphism and hypertension.
