摘要
目的:研究维通注射剂(Weitong injection,WTI)对大鼠腹膜正常成纤维细胞(normal fibroblast,NFB)与粘连成纤维细胞(adhesive fibroblast,AFB)增殖活性与细胞周期的影响。方法:利用原代培养获得大鼠NFB与AFB,以地塞米松为阳性对照药,采用MTT法和流式细胞术考察WTI对2种成纤维细胞(fibro-blast,FB)增殖活性与细胞周期的影响。结果:WTI对AFB的半数抑制浓度(50%inhibitory concentration,IC50)显著低于NFB(P<0.01),WTI对2种FB的IC50均显著低于地塞米松(P<0.01);WTI高、中剂量组和地塞米松组的AFB G0/G1期比例显著高于AFB空白对照组(P<0.01),S期比例显著低于AFB空白对照组(P<0.01);WTI高、中剂量组AFB的G0/G1期比例显著高于地塞米松组(P<0.01);各药物组的NFB细胞周期与空白对照组相比无显著性差异。结论:WTI通过将AFB停滞于G0与G1期,延缓其进入S期,抑制AFB增殖活性,WTI对AFB的作用强于NFB。
Objective: To study the effect of Weitong injection(WTI) on proliferation and cell cycle of cultured fibroblasts from normal and adhesive peritoneal tissues in rats.Methods: Normal fibroblasts(NFB) and adhesive fibroblasts(AFB) of rats were harvested from primary cultures.MTT and flow cytometry were used to detect the effect of WTI on proliferation and cell cycle of NFB and AFB.Dexamethasone was used as a positive control.Results: The 50% inhibitory concentration(IC50) of WTI was significantly lower in AFB than in NFB(P〈0.01).The IC50 of WTI in both fibroblasts was significantly lower than that of dexamethasone(P〈0.01).The G0/G1 stage proportion of AFB was significantly increased by WTI and dexamethasone(P〈0.01).The S stage proportion was significantly reduced by WTI in AFB(P〈0.01).The G0/G1 stage proportion of AFB was significantly higher in WTI groups than in dexamethasone group(P〈0.01).There were no significant difference in cell cycle of NFB between treatment groups and control group.Conclusion: WTI can arrest AFB in G0 and G1 stage,delay AFB to enter S stage,and restrain the proliferation of AFB.Moreover,the effect of WTI is more sensitive on AFB than on NFB.
