摘要
目的:以胶原诱导性关节炎(CIA)大鼠为模型,观察洛伐他汀对CIA大鼠血清细胞因子的影响,为探索炎性关节炎新的治疗药物提供理论基础。方法:利用牛Ⅱ型胶原建立Wistar大鼠CIA模型,造模成功后随机分为空白对照组、CIA模型组、布洛芬组、甲氨蝶呤(MTX)组和洛伐他汀组。分别在第2周和第5周时用ELISA方法检测各组大鼠血清的肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、白介素8(IL-8)和干扰素诱生蛋白-10(IP-10)的水平;用实时荧光定量PCR方法检测第5周时大鼠白介素1β(IL-1β)和白介素17(IL-17)mRNA的表达量。结果:免疫后第5周,洛伐他汀组、布洛芬组和MTX组大鼠的血清TNF-α,IL-6和IP-10的水平显著低于CIA组,而洛伐他汀组和MTX组的IL-8水平与CIA组大鼠无显著差异。洛伐他汀组和MTX组的IL-1β和IL-17 mRNA的表达量显著低于CIA组。结论:洛伐他汀能抑制TNF-α,IL-6,IL-1β,IL-17和IP-10的水平。
Objective: To evaluate effect of lovastatin on serum cytokines in rats with collagen-induced arthritis(CIA) and to supply theoretical basis for exploring new treatments of inflammatory arthritis.Methods: Arthritis was induced by injecting bovine Ⅱ collagen(BIIC) into Wistar rats.The rats were randomly divided into five groups: normal control,CIA control,ibuprofen,methotrexate(MTX) and lovastatin groups.The effects of these agents were assessed by measuring the changes in serum cytokine levels including TNF-α,IL-6,IL-8 and IP-10.The expressions of IL-1β and IL-17 mRNA were detected by QRT-PCR on 5th week.Results: The serum levels of TNF-α,IL-6 and IP-10 in lovastatin group were decreased on 5th week.The expressions of IL-1β and IL-17 mRNA in lovastatin group were less than that of the CIA control group.Conclusion: Lovastatin can decrease serum levels and the expression of TNF-α,IL-1β,IL-6,IP-10,IL-17,MCP-1 and RANTES in rats with CIA.
