己酮可可碱通过抑制肿瘤坏死因子α发挥抗炎作用 被引量:1

摘要 目的:探讨己酮可可碱(PTX)是否通过对血清肿瘤坏死因子α(TNF-α)的调节而发挥对大鼠胶原诱导性关节炎(CIA)的保护作用。方法:建立大鼠CIA模型后随机分为正常对照组、生理盐水CIA模型组、PTX治疗组。分别观察各组大鼠的体重、后足容积,并对关节炎的肿胀程度进行评分;用ELISA检测不同时间点各组血清的TNF-α水平。结果:与CIA组相比,PTX治疗组大鼠的炎症得到了较好的控制。第48天时PTX组的TNF-α水平与正常对照组的TNF-α水平没有统计学差异(P>0.05),但与CIA组相比显著降低(P<0.05)。结论:PTX对大鼠CIA关节炎具有缓解症状、抑制炎症肿胀的作用,PTX能够显著降低血清TNF-α水平。PTX可能是通过抑制血清TNF-α水平来发挥抗炎作用。 Objective: To explore whether pentoxifyline(PTX) has the protection on the treatment of collagen-induced arthritis in rats by inhibiting tumor necrosis factor-α. Methods: Wistar rat arthritis was induced by bovine II collagen (BIIC). The rats were randomly divided into three groups including normal control group, CIA group treated with normal saline and PTX group. The body weight, the hind paw volumes, the arthritic index of all rats at different time points were observed and measured. The serum TNF-α level of every group at different time point was detected by ELISA. Results: There was no difference be- tween PTX group and normal control group (P 〉0.05) at the 48'h day, and the TNF-α level of PTX group was less than that of CIA group (P 〈 0.05) at the 48'h day. Conclusion: Pentoxifyline significantly ameliorates the symptoms of CIA rats, and inhibits the development of inflammatory swelling. PTX can significantly decrease the serum TNF-α level. PTX may play role of anti- inflammation by inhibiting the serum TNF-α level
