摘要
目的模拟临床上血糖控制不佳出现的糖浓度波动,研究波动性细胞外高糖浓度(FEHG)对肾小球系膜细胞(MsC)增殖及其纤维连接蛋白(Fn)合成的影响。方法利用四甲基偶氮唑蓝(MIT)比色法和竞争抑制ELISA法动态观察不同葡萄糖浓度及波动性高糖浓度对AsC增殖和FN合成的作用。结果高糖浓度抑制MsC增殖,此抑制作用和糖浓度成正相关,同时刺激MsC的FN合成增加;FEHG也抑制MsC的增殖,且对FN的合成有较强的刺激作用。结论细胞外高葡萄糖环境抑制MsC增殖,且呈浓度依赖性;波动性细胞外高葡萄糖环境抑制MsC增殖,且抗增殖作用大于持续性高葡萄糖浓度组;细胞外高葡萄糖环境和波动性细胞外高葡萄糖均增加MsCFn的合成,其作用机制尚待进一步研究,波动性高糖浓度对MsC的Fn合成的刺激作用可能在治疗不充分的糖尿病(DM)患者出现糖尿病肾病(DN)起重要作用。
Objective: In Order to observe the effects of hyperlgycemia on Mesangial cell(MsC). The effects of fluctuating extracellular high glucose(FEHG) on the proliferation and fibronectin (Fn) synthesis were studied. Methods:The Fn level in supernatant of culturedd MsC and MsC Proliferation were assessed by means of competitive inhibiting ELISA method and MTT colormetric method respectively. Results: High glucose levls inhibited the MsC proliferation and FEHG had the same inhibition effect. the Fn synthesis in FEHG was more higher than that in high glucose level. Conclusion: Fluctuating changes in extracellular glucose concentration may play an important pathological role for FN synthesis of MsC in patients with diabetes mellitus treated inadequatly.
