犬冠状静脉窦阻塞后心肌肌钙蛋白Ⅰ和纤维连接蛋白免疫组织化学的研究

摘要 目的探讨冠状静脉窦阻塞后,对心肌组织肌钙蛋白Ⅰ(cTnI)和纤维连接蛋白(Fn)的影响。方法健康家犬18只,随机分为对照组、阻塞1 h组和阻塞2 h组,每组6只,对照组不结扎冠状静脉窦,后2组从心外膜在最近冠状静脉窦口处,结扎冠状窦,分别于阻塞后1 h和2 h处死犬。3组均在左心室前壁同一部位取材,行免疫组织化学染色,并进行分析。结果 3组犬实验前后血浆cTnI和Fn含量无明显变化(P>0.05)。与对照组比较,阻塞1 h组和阻塞2 h组心肌组织cTnI表达明显下降,Fn表达明显升高(P<0.01),且阻塞2 h组较阻塞1 h组变化更明显(P<0.01)。结论健康犬冠状静脉窦阻塞后,可导致心肌组织cTnI、Fn呈缺血性改变。 Objective To investigate the effect of coronary vein sinus occlusion on myocardial troponin I(cTnI) and fibronectin(Fn). Methods Eighteen healthy dogs were divided into control, block 1 hour and block 2 hours groups randomly. All dogs received chest open operation and coronary vein sinus was ligated at the site nearest the orifice of coronary vein sinus from epieardium. The animals were sacrificed after an hour or two hours respectively. In the control group,coronary vein sinus was not ligated. Then their hearts were taken out and samples of left ventricle were taken at same loeation. The immunohistochemical method was used to examine the expression of cTnI and Fn. Results Expression of cTnI and Fn of myocardium showed no significant diference after experiment compared with before. Compared with control group, expression of eTnI in block 1 hour and block 2 hours groups significanlly decreased, while expression of Fn significantly in- creased,and the changes were more significant in bloek 2 hours group than in block 1 hour group (P 〈0.01). Conclusion Coronary vein sinus occlusion caused myocardial ischemic injury.
