摘要
目的:比较和分析心理应激与复合应激模型大鼠行为和脑组织基因表达差异。方法:测定心理应激与复合应激模型大鼠负重游泳力竭时间和游泳不动时间;提取大鼠脑组织mRNA与基因芯片杂交,扫描并转换数据,分析和比较基因表达变化的差异。结果:与正常对照组相比,两种应激模型大鼠游泳不动时间均显著延长,与复合应激模型大鼠相比,心理应激模型大鼠游泳不动时间更长;两种应激类型大鼠脑组织基因表达发生显著改变的基因有132条,其中:下调基因有81条相同,上调基因20条相同。结论:心理应激与复合应激都能显著降低大鼠主观驱动性;心理应激与复合应激对大鼠脑组织基因表达的影响作用相似,心理应激的影响更为显著和广泛。
Aim: To compare and analyze the differences of the ethological pattern and brain tissue gene expression between psychological stress and combined stress in rat.Method: To determinate the loading swim time(LST) and forced swim time(FST) of both model groups;The mRNA which to be extracted from the brain tissue of both model group rats,were hybridized with Oligo microarray,and were screened through the analysis of gene expression profile,and to compare and analyze the change of gene expression.Results: The FSTs were extended in both model groups significantly.Compared with combined stress group,the FST were longer in psychological stress group.Totally,132 genes of the brain tissue were influenced in both groups,where the 81 down-regulated genes and 20 up-regulated genes were the same in both groups.Conclusion: Both types of stress can reduce the psychological impulse,and have similar influence on the gene expression of brain tissue in rat,the effects of psychological stress are more significant and wide-ranging.
