摘要
用氧化钙和氧化镁结合对铬革屑进行碱性处理提取明胶,然后用1398中性蛋白酶水解碱处理过的残渣提取得到蛋白质多肽。其较佳的工艺方法为,氧化钙与氧化镁的用量分别均为3%(以革屑干基质量计),水用量为300%,碱水解温度80℃,时间为4h,明胶的提取率在40%以上;酶用量0.125%,酶水解温度为46℃,蛋白多肽的提取率可达到20%;获得明胶和蛋白质多肽中CrO含量均低于50mg/kg,灰分含量均低于2%。
Gelatin was extracted from chrome shavings with alkaline treatment using CaO and MgO together.Then protein polypeptide was prepared by hydrolyzing the residue,which was left in the first step,with 1398 neutral proteinase.The optimum condition is as follows:the usage of CaO is 3%(based on the dry weight of chrome shavings),MgO is 3%,H2O is 300%,hydrolysis temperature is 80℃,and the reaction time is 4h,the yield of the gelatin is more than 40%.When the quantity of the enzymes is 0.125%,and the hydrolysis temperature is 46℃,the yield of the protein polypeptide can reach 20%.The quantity of Cr2O3 in both gelatin and protein polypeptide are lower than 50mg/kg,and the content of ashes is lower than 2%.
