摘要
目的：建立高效液相色谱法测定七味广枣丸中木香烃内酯和去氢木香内酯含量。方法：采用Agilent ZORBAX EclipseSB-C18（4.6 mm×150 mm,5μm）色谱柱,流动相为5%乙腈-乙腈（58∶42）,流速1 mL.min-1,检测波长为225 nm。结果：木香烃内酯和去氢木香内酯的保留时间分别约为21 min和24 min,与相邻峰的分离度大于1.5;线性范围分别为7.400~185.0μg.mL-1（r=0.9999）和5.950~148.8μg.mL-1（r=0.9998）;平均回收率（n=6）分别为101.3%和97.0%,RSD均为1.6%。结论：该法操作简便,测定结果准确,重复性好,可用于七味广枣丸中木香烃内酯和去氢木香内酯的含量测定。
Objective：To establish an HPLC method for the determination of costunolide and dehydrocostus lactone in Qiwei Guangzao pills.Methods：Agilent ZORBAX Eclipse SB-C18（4.6 mm×150 mm,5 μm） column was adopted,the mobile phase consisted of 5% acetonitrile-acetonitrile（58∶42） at the flow rate of 1 mL·min-1,and the detection wavelength was set at 225 nm.Results：The retention time of costunolide and dehydrocostus lactone was about 21 min and 24 min,and the resolution was more than 1.5;The linear ranges of costunolide and dehydrocostus lactone were 7.400-185.0 μg·mL-1（r=0.9999） and 5.950-148.8 μg·mL-1（r=0.9998）,respectively;The average recoveries of costunolide and dehydrocostus lactone were 101.3% and 97.0%,and RSDs of both were 1.6%.Conclusion：This method is simple,time-saving and accurate.It can be used for routine analysis of costunolide and dehydrocostus lactone in Qiwei Guangzao pills.
