摘要
本文比较了纤维素酶+酸性蛋白酶(方案Ⅰ)和纤维素酶+果胶酶(方案Ⅱ)两种酶解方案提取苦瓜皂甙的效果,结果表明:方案Ⅱ在正交最佳条件下,提取物中的皂甙纯度达13.10%,多糖含量12.82%,皂甙与多糖的含量均较高,试验对比了以上酶水解提取法与乙醇法提取苦瓜的皂甙纯度及紫外吸收光谱图,发现醇提苦瓜的皂甙纯度为4.11%,明显低于酶提取法;3种提取物中都含有三萜类皂甙和甾体类皂甙,但是乙醇法提取物中甾体类皂甙比两种组合式酶水解法要少。
In this paper,two kinds of extraction method using compound enzymes were compared, that cellulose enzyme assembled with acid protease( method Ⅰ ) and cellulose enzyme assembled with pectinase( method Ⅱ). The result show that purity of saponins and polysaccharide were respectively 13.10% and 12.82% in the Momordica charantia extracts ( method Ⅱ). The purity of saponins and UV absorption in enzymatic extraction and ethanol extraction have been compared. The purity of saponin extracted by ethanol was 4.11%, which was lower than enzymatic extraction significantly. There are triterpenoid saponins and steroidal saponins in all of the extracts ,but steroidal saponins in the ethanol extracts was less than in enzymatic hydrolysis extracts.
