摘要
目的探讨天花粉蛋白对体内外单纯疱疹病毒-1(HSV-1)DNA聚合酶和DNA结合蛋白mRNA表达的抑制作用。方法 36只小鼠随机分为正常组、模型对照组和天花粉蛋白治疗组,每组各12只,小鼠颅内接种HSV-1建立病毒性脑炎模型,治疗组于接种HSV-1前30min腹腔注射天花粉蛋白注射液,于接种病毒后12h、24h、48h、96h分别测定脑组织HSV-1DNA聚合酶和DNA结合蛋白mRNA表达水平。体外培养神经细胞分为正常组、药物对照组、病毒对照组和药物预处理组,接种病毒后12h、24h、48h、96h分别测定各组培养神经细胞的HSV-1DNA聚合酶和DNA结合蛋白mRNA表达水平。结果天花粉蛋白治疗组于小鼠颅内接种HSV-1后12h、24h、48h、96h脑组织HSV-1DNA聚合酶mRNA表达水平(8.39±0.84、9.62±0.87、7.81±0.90、7.66±0.85)显著低于模型组(11.26±1.02、12.43±1.17、11.56±1.68、12.78±1.71)(P<0.05);治疗组脑组织HSV-1DNA结合蛋白mRNA表达水平(7.28±1.07、8.25±0.78、7.56±0.96、7.85±1.22)显著低于模型组(10.45±2.72、12.33±1.97、11.24±1.86、12.54±2.32)(P<0.01)。药物预处理组在12h、24h、48h、96h培养神经细胞HSV-1DNA聚合酶mRNA表达水平(9.33±0.87、9.57±0.97、9.73±0.94、9.84±0.86)显著低于病毒对照组(10.15±1.12、15.63±1.24、12.77±2.38、16.58±1.92)(P<0.05);药物预处理组在24h、48h、96h培养神经细胞HSV-1DNA结合蛋白mRNA表达水平(7.58±1.75、8.63±1.79、7.38±1.47)显著低于病毒对照组(10.24±2.16、10.85±1.67、11.63±2.05)(P<0.05)。结论天花粉蛋白对体内外HSV-1DNA聚合酶和DNA结合蛋白mRNA表达具有抑制作用。
Objective To evaluate the effects of Trichosanthin(TCS) on Herpes simplex virus-1 (HSV-1) DNA polyase and DNA combine protein mRNA expression in vivo and in vitro.Methods Thirtysix mice were randomly assigned into three groups:Norm control group(n=12) ;Model control group(model group,n=12) ;TCS-treated group(treated group,n=12).Viral encephalitis was induced by intracranial inoculation of HSV-1 in the two groups.The TCS-treated group was injected with TCS 30 min before HSV-1 inoculation.The HSV-1 DNA polyase and DNA combine protein mRNA expression were measured at 12 h,24 h,48 h,96 h after HSV-1 inoculation,respectively.The culture neuron in vitro were assigned into four groups:Norm control group;drug control group;HSV-1 control group;TCS-pretreated group.The HSV-1 DNA polyase and DNA combine protein mRNA expression were measured at 12 h,24 h,48 h,96 h after HSV-1 were inoculated into the neuron,respectively.Results The HSV-1 DNA polyase mRNA expression of brain tissue at 12 h,24 h,48 h,96 h after HSV-1 intracranial inoculation were markedly lower in the TCS-treated group(8.39 ± 0.84,9.62 ± 0.87,7.81 ± 0.90,7.66 ± 0.85) compared with the Model group(11.26 ± 1.02,12.43 ± 1.17,11.56 ± 1.68,12.78 ± 1.71),respectively(P 〈 0.05).The HSV-1 DNA combine protein mRNA expression of brain tissue at 12 h,24 h,48 h,96 h after HSV-1 intracranial inoculation were markedly lower in the TCS-treated group(7.28 ± 1.07,8.25 ± 0.78,7.56 ± 0.96,7.85 ± 1.22) compared with the Model group(10.45 ± 2.72,12.33 ± 1.97,11.24 ± 1.86,12.54 ± 2.32),respectively(P 〈 0.01).The HSV-1 DNA polyase mRNA expression of neuron at 12 h,24 h,48 h,96 h after HSV-1 inoculation in the TCS-pretreated group(9.33 ± 0.87,9.57 ± 0.97,9.73 ± 0.94,9.84 ± 0.86) were markedly lower than that in the HSV-1 control group(10.15 ± 1.12,15.63 ± 1.24,12.77 ± 2.38,16.58 ± 1.92),respectively (P 〈 0.05).The HSV-1 DNA combine protein mRNA expression of neuron at 24 h,48 h,96 h after HSV-1
