摘要
应用索氏提取法、酸性乙醚提取法、正己烷提取法和氯仿-甲醇法4种方法,提取山稻蝗中的脂类化合物,结果显示氯仿-甲醇法的提取率最高,达7.68%。通过正交试验比较不同的索氏提取时间、丙酮浸提温度、浸取时间和浸取剂浓度分离脂肪中磷脂的效果,确定了提取磷脂的最佳条件:索氏提取时间6h,丙酮浸提温度为30℃,丙酮浸提时间为3h,丙酮的浓度500mL/g,此条件下磷脂的提取率为5.76%。
Four methods, Soxhlet, acidic aether, n-hexane and chloroform-methanol, were used to extract lipid compounds from Oxya agavisa Tsai. Results showed that the chloroform-methanol method had the highest extraction ratio(7.68%). Furthermore, the phospholipid was isolated from lipid compounds. Orthogonal test was used to compare different extraction time of Soxhlet method, concentration of solvent, acetone leaching time and temperature for separating phospholipid from lipids. The optimized process of phospholipid extraction were, Soxhlet extraction time, 6h; acetone leaching temperature, 30℃; leaching time, 3h;acetone concentration, 500ml/g, the extracting ratio of phospholipid under this condition was 5.76%.
