摘要
目的 :研究新的桂皮酰胺类衍生物 3 ,4 -二氯苯丙烯酰另丁胺 (AED880 1)对大鼠脑缺血 /再灌注损伤的影响。方法 :采用Pulsinelli四血管阻断大鼠脑缺血 /再灌注模型 ,观察了AED880 1对脑组织中水含量、氨基酸含量、6-keto-PGF1α和TXB2 含量 ,以及二者之比值的影响。结果 :一AED880 1能够降低脑缺血 /再灌注大鼠所引起的脑水含量增加。二采用氨基酸自动分析仪进行检测 ,发现脑缺血 /再灌注可致大鼠脑组织中谷氨酸 (Glu)含量增加 ,AED880 1可以使其降低 ,1 50mg/kg作用非常显著 (P <0 .0 1 )。对所测其它氨基酸含量虽有所影响 ,但均无统计学意义。三AED880 1可增加脑缺血 /再灌注大鼠脑中 6-keto -PGF1α含量 ,减少TXB2 含量 ;因而增加二者之比值。结论 :AED880 1对脑缺血 /再灌注具有保护作用 ,这种作用可能与其降低脑中Glu含量 ,增加脑中PGI2 含量和减少TXA2 含量 。
OBJECTIVE:To investigate the protective effect of 3,4-dichloraphenyl-propenoyl-sec.-butylamine(AED 8801 ) on cerebral ischemia/reperfusion in rats and to study its protective mechanisms.METHOD:The model of cerebral ischemia/reperfusion in rats was established by the occlusion of four-vessel to study the protective effect of AED 8801 .The brain liquid content,the level of some amino acids and the levels of 6-keto-PGF 1α and TXB 2 were measured.RESULTS:AED 8801 50,100 and 150mg/kg ip can decrease the brain liquid content,AED 8801 150mg/kg has very significant effect(P<0.01).AED 8801 50,100 and 150mg/kg ip can reduce the level of Glu in brain tissue.AED 8801 can increase the level of 6-keto-PGF 1α (P<0.01) while decrease the level of TXB 2(P<0.05) in brain tissue,corresponding elevate the ratio of 6-keto-PGF 1α /TXB 2(P<0.01).CONCLUSION:AED 8801 can antagonize the experimental cerebral ischemia effectively.The protective effect may be due to decreasing Glu content in whole brain,increasing PGI 2 content while decreasing TXA 2 content in whole brain,and rasing the ratio of PGI 2/TXA 2.
