摘要
目的研究肿瘤干细胞标记物CD133和CD44在肺癌原发灶及转移淋巴结中的表达情况,及其与肿瘤生物学特征的关系。方法采用免疫组化法检测CD133和CD44在42例肺癌患者的肿瘤原发灶及转移淋巴结中的表达,并分析其表达情况与肿瘤大小、组织学分型、细胞分化程度、淋巴结转移情况的关系。结果 42例肺癌组织中,淋巴结转移占64.3%(27/42)。原发灶CD133阳性率73.8%(31/42);CD44阳性率59.5%(25/42)。转移淋巴结CD133阳性率92.6%(25/27),CD44阳性率81.5%(22/27)。CD133阳性组淋巴结转移率为74.2%(23/31),较阴性组的27.3%(3/11)高。CD44阳性组淋巴结转移率为76.0%(20/25),较阴性组的41.2%(7/17)高。原发灶CD133和CD44表达与转移淋巴结CD133和CD44表达呈正相关。肺癌组织中CD133与CD44的表达之间呈正相关性。CD133阳性组患者3 a生存率明显低于CD133阴性组(P<0.05)。CD44阳性组患者3 a生存率明显低于CD44阴性组(P<0.05)。结论肿瘤干细胞标志物CD133和CD44在肺癌的原发灶及转移淋巴结中的表达状态与肿瘤生物学特征密切相关,并影响患者的预后,对分析和预测肺癌临床表现具有较为重要的意义。
Objective To study the expression of tumor stem cell marker CD133 and CD44 in original tumor tissues and metastatic lymph nodes of lung cancer,and to explore their relationships with tumorous biological features.Methods The expression of CD133 and CD44 in original tumor tissues and metastatic lymph nodes of 42 patients with lung cancer was detected by immunohistochemical method,respectively.Then the relationships between their expression and some tumorous biological features such as tumor size,histological type,cellular differentiation degree,lymph node metastasis were analyzed.Results In total 42 patients,64.3%(27/42)of them had lymph node metastasis.In original tumors,the positive rate of CD133 and CD44 expression was 73.8%(31/42)and 59.5%(25/42),respectively,while in metastatic lymph nodes it was 92.6%(25/27)and 81.5%(22/27),respectively.The metastatic rate of lymph nodes was 74.2%(23/31)in CD133 positive group and higher than 27.3%(3/11)in CD133 negative group.The metastasis rate of lymph nodes in CD44 positive group was 76.0%(20/25)and also higher than 41.2%(7/17)in CD44 negative group.The expression of CD133 and CD44 in original tumors was positively correlated with their expression in metastatic lymph nodes,respectively.Moreover,CD133 expression was positively correlated with CD44 expression in lung cancer tissues.The 3-year survival rates both in CD133 and CD44 positive groups were apparently lower than those in CD133 and CD44 negative groups(P0.05).Conclusion The expression of tumor stem cell marker CD133 and CD44 is closely correlated with tumorous biological features,and they may affect the prognosis of patients with lung cancer.Therefore,they are important markers for the analysis and prediction of clinical behavior of lung cancer.
