摘要
目的观察孕鼠在强迫游泳实验(FST)中的绝望状态,检测FST后孕鼠海马区雌激素α受体(ERα)的表达,初步探讨孕鼠FST时绝望状态与海马区ERα水平的关系。方法同批次雌昆明鼠,包括28只孕鼠和20只对照鼠;进行6次FST,记录第2次和第6次的不动时间。FST后小鼠大脑标本以海马为中心作冠状面切片,免疫组化SP法检测ERα的表达。结果孕鼠在第2次和第6次FST中不动时间(分别为149.21±56.10 s、178.54±35.54 s)均长于非孕鼠(118.80±52.72s、150.15±37.92s),P分别为0.024、0.011。孕鼠和对照雌鼠海马区均有ERα表达,但孕鼠ERα组化评分(3.86±2.69)高于非孕鼠(1.54±1.85),P为0.008。小鼠海马区ERα免疫组化评分与第2次FST、第6次FST的不动时间均有直线相关,r值分别为0.332、0.394,P值分别为0.021、0.006。结论孕鼠在FST中比非孕鼠表现出更明显的绝望状态;其海马区ERα表达水平亦增高,可能与妊娠期抑郁有关。
Objective To observe the desperate state of pregnant mice in forced swimming test(FST)and detect the expression of estrogen receptor α(ERα)in the hippocampus of mice after swimming,and to study the relationship between the desperate state of pregnant mice in FST and the expression of ERα in hippocampus.Methods Female Kunming mice of the same batch included 28 pregnant mice and 20 control mice.FST was repeated for 6 times,and immobility durations in the second and the sixth FST were recorded.After FST,the cerebrum was sampled,and the hippocampus was the center of paraffin-embedded section.The expression of ERα was detected by immunohistochemistry.Results The immobility durations in the second and the sixth FST were significantly longer in preganant mice than in control mice(149.21±56.10 vs 118.80±52.72 s,P =0.024;178.54±35.54 vs 150.15±37.92 s,P =0.011),respectively.ERα were expressed in the hippocampus of both pregnant mice and control mice,and the immunohistochemical score of ERα in pregnant mice was signficantly higher than that in control mice(3.86±2.69 vs 1.54±2.07,P =0.008).There were linear correlations between the immunohistochemical score and the immoblity durations in the second and sixth FST(r =0.332,P =0.021;r =0.394,P =0.006),respectively.Conclusion Pregnant mice are more likely to show desperation in FST,and the up-regulated expression of ERα in the hippocampus of pregnant mice may relate to antepartum depression.
