温度敏感性脂质体磁性阿霉素纳米粒的制备 被引量:1

摘要 目的制备能够携带化疗药物阿霉素的温度敏感性脂质体磁性纳米粒,并对纳米粒的粒径大小、药物包裹率、稳定性等物理性质进行检测。方法以化学共沉淀法制备四氧化三铁纳米粒,以逆相蒸发法制备空白温度敏感性磁性脂质体纳米粒,梯度反向装载法包封阿霉素,激光粒度分析仪检测粒径大小分布范围,透射电镜观察粒子形态;酸性乙醇荧光分光光度法测量阿霉素包裹率和泄漏率。结果制备的温度敏感性脂质体磁性纳米粒平均粒径为(76±18)nm,粒径较均匀,平均包裹率为(42.6±0.7)%,室温下贮存稳定。结论本实验制备的温度敏感性脂质体磁性纳米粒基本符合要求,可为进一步研究提供了基础。 【Objective】 To prepare thermo-sensitive-liposome-magneticadriamycin-nanoparticles (TsLMADM), measure physical property of the Ts-LMADM, such as size of nanospheres, magnetism, content of ADM, drug loading (DL) and stability. 【Methods】Prepared four iron oxide nanoparticles by chemical coprecipitation method, prepared blank thermo-sensitive-liposome-magnetic nanoparticles by reverse-phase evaporation method, then encapsulated adriamycin by reverse gradient loading method. Detected the distribution range of the particles' size by laser particle size analyzer, observed particles' morphology by transmission electron microscopy, measure entrapment efficiency and leakage rate of adriamycin by acidic ethanol fluorescence spectrophotometry.【Results】Average size of thermo-sensitive-liposome-magnetic nanoparticles prepared is (76 ± 18) nm, the particle size range is narrow, entrapment efficiency is (42.6 ± 0.7) %, particles show stability at room temperature.【Conclusion】In this study, the thermo-sensitive-liposome-magnetic nanoparticles basically meet the requirements, and can provide the basis for further study.
