摘要
目的:建立测定保健食品中大豆低聚糖(包括蔗糖、棉子糖、水苏糖3种成分)的高效液相色谱含量测定方法。方法:色谱柱Agilentzorbax NH2(4.6mm×250mm,5μm),柱温25℃;流动相水-乙腈(30∶70),流速1.2mL·min-1;示差折光检测器,内部温度35℃。结果:蔗糖、棉子糖、水苏糖的线性范围分别为2.38~23.8μg,2.04~20.4μg,2.24~22.4μg;加样回收率(n=5)分别为96.4%,97.5%,97.4%。结论:本方法简便、灵敏、准确,可用于含大豆低聚糖保健食品中以上3种成分的含量测定和产品质量控制。
Objective:To develop an RP-HPLC-RID method for determination of soybean oligosaccharide(including sucrose,raffinose,stachyose)in functional foods.Methods:An Agilent zorbax NH2 column (4.6 mm×250 mm,5 μm)was applied; a mobile phase composing of acetonitrile and water(70∶30)was used,and the flow rate was 1.2 mL·min^-1; A RID detector was applied,and the inner temperature was set at 35 ℃.Results:The results were obtained that quantitative line range of three oligosaccharide was 2.38-23.8 μg,2.04-20.4 μg,and 2.24-22.4 μg respectively,and the average recovery (n=5) was 96.38%,97.52%, and 97.40% respectively.Conclusion:The method is specific,sensitive,simple and reproducible ,and can be used for the determination of the three ingredients in functional foods that claimed containing soybean oligosaccharides.
