曲妥株单抗联合顺铂对卵巢癌细胞株的抑制作用 被引量:1

Inhibitory Effects of Trastuzumab Combined with DDP on Cell Line of Human Ovarian Cancer

摘要 目的研究曲妥株单抗对卵巢癌细胞株SKOV3的抑制作用。方法采用四甲基偶氮唑盐(MTT)法与流式细胞术检测人卵巢癌细胞株SKOV3的生长抑制及凋亡情况。结果 (1)顺铂(DDP)、曲妥株单抗及DDP+曲妥株单抗合药均对卵巢癌细胞株SKOV3有一定的生长抑制作用;在同一浓度、同一作用时间下,DDP组的抑制率高于曲妥株单抗组,DDP+曲妥株单抗组的抑制率高于DDP组;3组均随着药物浓度的增加、作用时间的延长而抑制作用增强(P<0.05)。(2)DDP、曲妥株单抗及DDP+曲妥株单抗3组的凋亡率均较对照组高,且3组的凋亡率均随作用时间的延长而升高;在同一作用时间下,DDP组的凋亡率高于曲妥株单抗组,DDP+曲妥株单抗组的凋亡率高于DDP组(P<0.05)。结论 DDP、曲妥株单抗及其联合DDP均能明显抑制人卵巢癌细胞株SKOV3的增殖,诱导细胞凋亡,并呈剂量和时间依赖性,联合DDP效应增强。 Objective To study the inhibitory effects of trastuzumab on ovarian cancer cell line SKOV3. Methods MTT assay and Flow cytometry was used to compare Cisplatin ( DDP) ,trastuzumab and DDP combined with trastuzumab on inhibiting the growth and apoptosis-inducing effect of high expression HER-2 /neu ovarian cancer cell line SKOV3. Results ( 1) In three groups,cell line SKOV3 was inhibited,and the effect increasd with drug concentration and drug action time. In the same concentration inhibition ratio of three groups raised with drug action time,and in the same drug action time inhibition ratio raised with drug concentration. In the same concentration and action time inhibition ratio of DDP was higher than that of trastuzumab,while that of DDP combined with trastuzumab was higher than that of DDP( P 0. 05) . ( 2) Apoptosis ratio of DDP,trastuzumab and DDP combined with trastuzumab was significantly high compared to the control. Apoptosis ratio in these three groups raised with drug action time. In the same time apoptosis ratio of DDP was higher than that of trastuzumab,while that of DDP combined with trastuzumab was higher than that of DDP ( P 0. 05) . Conclusion DDP,trastuzumab and DDP combined with Trastuzumab significantly inhibit cell line SKOV3 proliferation and induce cell apoptosis, and the effect indicated drug concentration and time-dependent. DDP combined with trastuzumab possesses obvious synergy.
