脑溢安颗粒对实验性脑出血大鼠转化生长因子-β1表达的影响

摘要 目的研究脑溢安颗粒对实验性脑出血大鼠脑内转化生长因子-β(1 TGF-β1)mRNA和蛋白表达的影响。方法通过立体定位向脑内苍白球注入Ⅶ型胶原酶0.4U建立脑出血模型,运用原位杂交法和Westernblotting检测脑出血后不同时间点TGF-β1的表达变化。结果 TGF-β1在脑出血3h已明显表达,6h进一步增强,但在12h表达下降,而在1d后第2次升高达到高峰,4~7d减少,14d后消失。脑溢安组TGF-β1表达在6h、1d、4d显著高于模型组。结论脑出血后脑内TGF-β1表达增加;脑溢安可上调脑出血后TGF-β1的表达,可能是其神经保护机制之一。 Objective: To study the Naoyi’ an Granule on expressions including mRNA and protein of transforming growth factor-β1(TGF-β1) in experimental rats with cerebral hemorrhage.Methods: The intracerebral hemorrhage(ICH) animal models were made by injecting collagenase type Ⅶstereotaxically into right globus pallidus.The TGF-β1 expressions in servral time points were detected by the hybridization in situ method and the western method.Results: The TGF-β1 expression was obvious in the 3h after cerebral hemorrhage.In the 6th hour,this expression was more remarkable.However,the index was decreased in the 12th hour.In the 1st day,the TGF-β1 was increased the top level.From the 4th day to 7th day,it was reduced.Until the 14th day,it was disappeared.In the 6th hour,the 1st day and the 4th day,the TGF-β1 expression in the Naoyi’ an Granule group was higher than that in the model group.Conclusion: The TGF-β1 expression is increased after cerebral hemorrhage.That this index is increased by the Naoyi’ an Granule may be one of the mechanisms of neuroprotective effect.
