摘要
目的通过观察补阳还五汤对脑出血大鼠脑内损伤区血管内皮生长因子(vascular endothelial growth factor,VEGF)mRNA表达的影响,探讨补阳还五汤的作用机制。方法以立体定位向脑内苍白球注入Ⅶ型胶原酶0.5U,建立脑出血模型。95只SD大鼠随机分为正常组5只、假手术组、模型组、补阳还五汤(BYHWD)组各30只,BYH-WD组灌服补阳还五汤,运用逆转录聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)检测正常组及其他组造模术后1,4,7,14,21,28d各时间点VEGF mRNA的动态变化。结果正常组及假手术组不同时间点VEGF mRNA未见明显表达;模型组、补阳还五汤组在脑出血术后1d即有表达,其后逐渐增多,模型组在术后21d达到高峰(P<0.01);而补阳还五汤组在术后14d即达到高峰(P<0.01),随后开始下降;从第7天开始补阳还五汤组即高于模型组(P<0.05)。结论补阳还五汤能促进脑出血大鼠脑内VEGF mRNA的表达,从而调整血管新生过程,这可能是其促进组织修复的机制之一。
Objective To investigate the mechanisms of Buyang Huanwu Decoction (BYHWD) through observing its effects on expressions of vascular endothelial growth factor(VEGF) mRNA in cerebral hemorrhagic(CH) rat brains. Methods CH model was induced by stereotaxical injection of collagenase type Ⅷ into right globus pallidus. Ninety five Sprague - Dawley rats were randomly divided into four groups: normal control group( n = 5), sham group( n = 30), ICH model group( n = 30), and BYHWD - treated group( n = 30). Animals in BYHWD - treated group were administered orally with BYHWD. The expression of VEGF mRNA was assayed by reverse transcription- polymerase chain reaction (RT - PCR ) 1, 4, 7, 14, 21 and 28 day(s) after the modeling. Results The expression of VEGF mRNA in rat brains of normal and sham groups was not obvious. The VEGF mRNA expression occured in rat brains at the first day after the onset of CH, then increased gradually, and arrived the peak at 14th day( P 〈 0.01) in BYHWD group and at 21st day ( P 〈 0.01) in CH model group, then all decreased till to the 28 th day. Expressions of VEGF mRNA in BYHWD - treated group were significantly higher than those in CH model group from 7 days after onset to 28 d ( P 〈 0.05). Conclusion BYHWD can promote the expressions of VEGF mRNA in cerebral hemorrhagic rats, which may be related with the enhancement of the reconstruction of the microvascular networks in the damaged zone.
