摘要
目的探讨大鼠脑缺血再灌注损伤后Survivin和Caspase-3表达与细胞凋亡的关系。方法将42只成年雄性SD大鼠,体重250~280g,随机分成对照组(n=3)、假手术组(n=3)和缺血再灌注6h、12h、1d、2d、3d和6d组(n=6)。采用栓线法制备大脑中动脉缺血90min再灌注模型,应用免疫组织化学方法观察Survivin和Caspase-3表达,TUNEL方法检测细胞凋亡。结果对照组及假手术组未见Survivin和Caspase-3表达,仅见凋亡细胞0~2个,缺血再灌注6h,显示在缺血侧皮层、皮层下纹状体和海马区Survivin和Caspase-3表达增多,凋亡细胞增多,12h~1d三者均持续增多,2d Caspase-3表达及凋亡细胞达到高峰,3~6d呈下降趋势,而Sur-vivin的表达3d达高峰,持续至6d有下降趋势(P<0.05)。结论大鼠局灶脑缺血再灌注损伤后诱发Survivin和Caspase-3异常表达,并与细胞凋亡有密切联系。
Objective To study the cerebral ischemia reperfusion injury Survivin and Caspase-3 expression and cell apoptosis.Methods42 adult male SD rats weighing 250~280 g, were randomly divided into control group(n = 3), sham operation group(n =3), ischemia-reperfusion 6 h, 12 h, 1 d, 2 d, 3 d, 6 d group(n =6). Prepared by the filament 90 minutes of middle cerebral artery occlusion and reperfusion model was observed by immunohistochemistry Survivin and Caspase-3 expression, TUNEL to detect apoptosis. ResultsIn the control group and the sham group no Survivin and Caspase-3 expression seen in apoptotic cells from 0 to 2, ischemia-reperfusion 6 h, displayed in the ischemic cortex, striatum and hippocampus subcortical Survivin and Caspase-3 increased expression, and apoptosis of cells increased, 12 h 1 d none of the three continued to increase, 2 d Caspase-3 expression and apoptosis peak, 3~ 6 d downward trend, while the expression of Survivin 3 d peak, continuing to 6 d a downward trend.Conclusion Focal cerebral ischemia and reperfusion injury induced by Survivin and abnormal expression of Caspase-3, and is closely related with cell apoptosis.
