葛根素对百草枯诱导大鼠肺纤维化转化生长因子β1的影响 被引量:1

摘要 [目的]探讨葛根素(puerarin)对百草枯(paraquat,PQ)致肺纤维化的治疗作用,及其对大鼠肺组织转化生长因子β1(transforming growth factor-β1,TGF—β1)的影响。[方法]复制大鼠百草枯致肺纤维化模型,光镜观察组织学改变,测定血浆中超氧化物歧化酶(SOD)活性、丙二醛(MDA)和肺组织均浆中羟脯氨酸(HYP)和转化生长因子β1(TGF—β1)含量。[结果]PQ染毒后血浆中超氧化物歧化酶(SOD)降低,血浆丙二醛(MDA)和肺组织均浆中HYP和TGF-β1含量升高,与正常对照组相比,相应观察时间点差异有统计学意义(P〈0.01)。葛根素能降低PQ致肺纤维化大鼠肺组织中过高的HYP和TGF—β1水平和血浆中MDA的含量,提高大鼠体内的SOD的活性。病理组织学检查亦表明,葛根素明显改善PQ致大鼠肺纤维化程度。[结论]葛根素对百草枯致肺纤维化具有一定的治疗作用,其作用机制可能与降低大鼠肺组织TGF—β1水平有关。 [ Objective] To investigate the curative effect of puerarin on pulmonary fibrosis in rats induced by paraquat, and its influence on transforming growth factor- β1 (TGF- β1). [Methods] Model of pulmonary fibrosis in rats was induced by paraquat and they were treated with puerarin. The histological changes were observed with microscope. The level of maleic dialdehyde (MDA),and the activity of superoxide dismutase (SOD) in plasma , as well as the content of hydroxyproline(HYP) and the levels of TGF-β 1 in pulmonary tissue were determined.[Resuhs] The level of MDA in plasma and levels of HYP and TGF- β1 in pulmonary tissue increased and the activity of SOD in plasma decreased with positive significance compared with control group (P〈0.01). Puerarin significantly reduced the elevate levels of HYP and TGF- β 1 in pulmonary tissue and MDA in plasma, and increase the activities of SOD in plasma. Pathological examination suggested puerarin has the rapeutic effect on experimental pulmonary fibrosis with significant. [Conclusion] Puerarin may have the rapeutic effect on paraquat-induced pulmonary fibrosis in rats, and the mechanism of its action may be related to down-regulating TGF- β1 in pulmonary tissue.
