摘要
对比研究了桑黄摇瓶和发酵罐培养不同阶段的各项生理指标.结果表明:摇瓶培养在10%接种量、28℃、160 r/min的条件下,菌丝量168 h达到最大,还原糖及氨基态氮分别在120 h和72 h达到最高峰,最适pH为5.5;发酵罐培养在5 L装液量、10%接种量、200 r/min搅拌速度、通气量1∶1(v/v)的条件下,菌丝量120 h达到最大,还原糖及氨基态氮含量随桑黄的生长呈下降趋势,最适发酵pH为5.5.可见发酵罐培养与摇瓶培养是有显著差异的,掌握发酵罐培养的规律,可以实现桑黄的大规模生产.
The paper compared Phellinus igniarius shake flask culture with fermenter culture at different stages of the physiological indices,it showed that: shake flask culture in conditions that the inoculum was 10%,the temperature was 28 ℃ and the speed was 160 r/min,make the mycelium volume in the 168 h reached maxmium,reducing sugars and amino nitrogen in the 120 h and 72 h respectively,reached a peak,and the optimal pH value was 5.5;fermenter culture in conditions that the liquid volume was 5 L,the inoculum was 10%,stirring speed was 200 r/min and ventilation was 1∶1(v/v),the mycelium volume in the 120 h reached the maxmium,reducing sugar and amino nitrogen content declined with the growth of Phellinus,and the optimum fermentation pH value was 5.5.There is significantly difference between fermenter culture and shake flask culture,and to master the law of fermenter culture can realized large-scale production of Phellinus igniarius.
