摘要
目的:建立复方痤疮微乳膏中阿奇霉素的萃取与测定方法。方法:精密称取复方痤疮微乳膏,精密加入三氯甲烷使溶解完全;精密加入0.1mol·L-1盐酸溶液,振荡5min,静置15min。定量吸取萃取液,用0.1mol·L-1盐酸溶液定容。精密量取5mL,与85%硫酸溶液5mL混合,振摇1min,室温下放置30min。分光光度法测定λ482nm处的吸光度。结果:阿奇霉素浓度在25~65μg·mL-1范围内线性相关性好(r=0.9996,n=5)。盐酸溶液萃取阿奇霉素的回收率为92.86%,精密度RSD为0.91%(n=9)。制剂中其他成分和辅料不干扰测定,样品测定重现性好(RSD=0.89%,n=5)。结论:该方法简单、快速、经济,适用于复方痤疮微乳膏中阿奇霉素的质量控制。
Objective:To establish the method for extraction and determination of azithromycin in compound acne microemulsion cream.Methods:Compound acne micro-emulsion cream was precisely weighed and then completely dissolved in chloroform with precise amount.Azithromycin was extracted by precisely adding 0.1 mol·L-1 hydrochloric acid.After being shaked for 5 minutes,the mixture was settled for 15 minutes further.A certain amount of the top layer was extracted.The extraction was then diluted with 0.1 mol·L-1 hydrochloric acid.Five milliliter of the dilution was mixed with 5 mL of 85% sulfuric acid solution.The new mixture was shaken for 1 minute and settled for 30 minutes at room temperature.The absorbance at λ 482 nm was determined.Results:The concentration of azithromycin and the absorbance in the range of 25 ~ 65 μg·mL-1 had a strong positive correlation(r = 0.999 6,n = 5).The recovery of azithromycin in hydrochloric acid extraction was 92.86%,the RSD value was 0.91%(n = 9).The preparation of other components did not interfere with the determination,and determination of sample had a good reproducibility(RSD = 0.89%,n = 5).Conclusion:This extraction and determination method is simple,fast,and economical.It is suitable to be applied to the quality control of azithromycin in compound acne micro-emulsion cream.
