摘要
采用平板初筛和摇瓶复筛等方法,从秦皇岛渤海海域的海水样品中,分离获得产淀粉酶活力较高的菌株HYM-7,初步鉴定为解淀粉类芽孢杆菌(Paenibacillus.amylolyticus),设计单因素和正交实验,确定该菌的最佳发酵条件。结果表明:在装液量50mL液体培养基的250mL三角瓶中,采用单因素实验得最佳接种量为1%,最适产酶时间为48h,最适碳源为玉米淀粉,氮源为大豆粉,Ca2+和Mg2+可显著提高产酶量;采用正交实验得最适发酵培养基配方(g/L):玉米淀粉4、大豆粉10、可溶性淀粉2、MgSO40.2、CaCl20.4。对优化的发酵培养基进行验证,测得酶活力为34.62U·mL-1,较基础培养基酶活提高60.8%。
Through preliminary petri-dish and following by shaking flask fermentation screening, a highest-yield amylase HYM-7 was isolated from samples in Bohai Sea area-Qinhuangdao, and it was primarily identified as Paenibacillus.amylolyticus. Its fermentation condition was studied through the single- factor and orthogonal experiment.The results demonstrated that after 48h it had the maximum amylase activity under 1% the vaccination quantity when 50mL broth in 250mL flask,And the optimal carbon and nitrogen source was corn flour and soybean flour respectively, Ca^2+ and Mg2. were beneficial to enzyme activity obviously. Through orthogonal experiment obtained the top-seeded formula for the media(g/L) :corn flour 4,soybean flour 10, soluble starch 2, magnesium sulfate 0.2 and calcium chloride 0.4.Finally,the optimized fermentation medium was validated,and the total activity of amylase was 34.62 U · mL^-1 ,which was 60.8% more than basic medium.
