摘要
分别采用酶解法、CTAB法、SDS法、CTAB/NaCl法等4种方法提取金黄色葡萄球菌基因组DNA,并进行比较改良。结果表明,SDS法提取的DNA含量最高;当金黄色葡萄球菌的DNA质量稀释至1.975×10-4pg时,SDS法提取的DNA仍能用于PCR扩增;此外,该方法稳定性好、经济性高,是理想的金黄色葡萄球菌基因组DNA提取方法。
To devise a rapid and effective economic extraction of Staphylococcus Aureus'DNA,Four methods,included Lysozyme,CTAB,SDS and CTAB/NaCl were used to extract the DNA from Staphylococcus aureus,then used colorimetry、agarose gel electrophoresis and PCR to detect the extractives and compare the results.The result showed that the quality of DNA extracted by SDS method was the most amoung the four methods;when the quality of Staphylococcus Aureus' DNA diluted to 1.975×10-4 pg,SDS method can still detect the extractives by PCR。 SDS method was stable and economic and was an ideal extraction of Staphylococcus Aureus'DNA.
