摘要
目的利用近红外光谱法快速测定舒血宁注射液中总黄酮醇苷的含量。方法采集舒血宁注射液NIR光谱,采用偏最小二乘法(PLS)进行回归,经内部交叉验证建立舒血宁注射液的NIR光谱与其主要成分总黄酮醇苷的HPLC法测定值之间的多元校正模型,预测总黄酮醇苷的含量。结果校正集样品浓度范围为0.760~0.890mg.mL-1。内部交叉验证决定系数(R2)为79.4,内部交叉验证均方差(RMSECV)为0.0154,外部验证均方差(RMSEP)为0.0233。结论本方法快速、简便,适用于药品的现场快检。
Objective To develop a novel method for fast determination of the total flavonol glycosides in Shuxuening Injection with NIR.Methods NIR diffuse reflectance spectra of the injection were collected.Partial least square(PLS) was utilized to build the calibration model with these spectra using HPLC as a reference method for determination of the total flavonol glycosides in Shuxuening Injection.Results The determination coefficients R2 was 79.4 as indicated from the cross validation.RMSECV was 0.0154.RMSEP obtained from the validation was 0.0233.Conclusion The method is simple and rapid.The NIR result can be used in the mobile lab.
