莪术醇抑制人结肠癌SW1116细胞增殖及相关机制的研究 被引量:8

摘要 观察莪术醇对人结肠癌SW1116细胞生长及肿瘤细胞凋亡的影响,以探讨其可能的机制。用不同浓度的莪术醇处理SW1116细胞,MTT检测莪术醇对SW1116细胞生长增殖的抑制作用;流式细胞术(FCM)检测细胞凋亡率;比色法测定Caspase-3酶活性;Western blot法检测Bcl-2、Bcl-XL的蛋白表达。莪术醇能以时间、剂量依赖性的关系抑制人结肠癌SW1116细胞的增殖,并诱导凋亡;莪术醇与SW1116细胞作用48h和72h后Caspase-3酶活性显著增强;同时,莪术醇处理SW1116细胞后,Bcl-2和Bcl-XL蛋白表达下调。说明莪术醇可抑制人结肠癌SW1116的增殖,诱导其凋亡,机制可能与调节Caspase-3、Bcl-2/Bcl-XL表达水平有关。 To investigate the inhibitory effects of curcumol on proliferation inhibition and apoptosis induction in human colon carcinoma SW1116 cells and its possible mechanism,the proliferation inhibition was measured by MTT assay.The flow cytometer was used to analyze the apoptosis rate.Colorimetric method was employed to detect the activity of caspase-3.The expression of Bc l—2 and Bc l—XL proteins were measured by Western blotting assay,SW1116 cells growth and induced their apoptosis in dose and concentration-dependent manners are curcumol inhibited.After treatment with curcumol for 48 hours or 72 hours,the activity levels of caspase-3 significant increased.Furthermore,the expression levels of Bc l—2 and Bc l—XL could be decreased by curcumol in SW1116 cells.It is conclused that curcumol can inhibit the growth and induce the apoptosis of SW1116 cells significantly,which may be related with regulating the expression levels of Caspase-3,Bc l—2 and Bc l—XL.
