摘要
甲基丙二酸半醛脱氢酶(MMSDH)是唯一需要CoA的一类醛脱氢酶.采用免培养-PCR技术结合基因组步移从宏基因组中成功分离一条MMSDH,获得的MMSDH在序列上具有醛脱氢酶的10个保守模块,以芽胞杆菌的MMSDH为模板预测其三维结构.研究证实采用免培养-PCR技术及基因组步移在微生物基因研究中具有重要的价值,且为新酶的筛选技术提供一条简单有效的途径.
Methylmalonic acid semialdehyde dehydrogenase (MMSDH) is unique type of aldehyde dehydrogenase which requires coenzyme A for the reaction. The paper has cloned a MMSDH gene from metagenomics by culture-independent PCR and genome walking. The putative amino acid sequence of MMSDH has ten conservative motifs of aldehyde dehydrogenase. The three-dimensional structure of new MMSDH is construed based on bacillus MMSDH. The research has verified the value of culture- independent PCR and genome walking in microbial diversity studies and provided a simple and effective way to develop of a new enzyme screening technology.
