摘要
目的应用钙载体和ＰＣ１２细胞株探讨胞浆钙离子与神经细胞凋亡之间的关系。方法水母发光蛋白导入法测量ＰＣ１２胞浆钙离子，细胞存活率监测细胞死亡，ＤＮＡ琼脂糖电泳证实细胞凋亡。结果钙载体（Ａ２３１８７）升高ＰＣ１２胞浆钙离子，降低细胞存活率，ＤＮＡ琼脂糖电泳显示典型的ＤＮＡ梯形带。结论胞浆钙离子升高激活内源性核酸内切酶，导致ＤＮＡ核小体间断裂，启动神经细胞凋亡。
Objective Relationship between neuronal apoptosis and changes in cytoplasmic calcium was studied using A23187,calcium ionophore,and PC12 cell line. Methods Changes in cytoplasmic calcium was measured with aequorinloaded method, neuronal death was shown in cell viability,DNA fragmentation was used as an apoptosis indicator. Results A23187 caused an increase in cytoplasmic calcium,loss of cell viability,and DNA fragmentation in PC12 cell line. Conclusion Increase in cytoplasmic calcium activates endonuclease(s) which causes DNA fragmentation at internucleosomal sites and neuronal apoptosis.
