摘要
目的探讨成纤维细胞激活蛋白(FAP)和基质金属蛋白酶-1(MMP-1)在小鼠肝纤维化组织中的表达和两者之间的关系,及其在肝纤维化形成过程中的作用机制。方法建立四氯化碳诱导的小鼠实验性肝纤维化模型,HE染色和Masson染色法判定肝纤维化程度,采用免疫组织化学染色法测定FAP、MMP-1在肝纤维化过程中的时序动态表达。结果模型组中FAP和MMP-1的表达部位和范围都一致,阳性表达主要分布于门管区内血管壁周围及纤维间隔内的成纤维细胞的胞浆中。线性相关分析显示FAP和MMP-1与小鼠肝纤维化程度呈正相关(r分别为0.672,0.684;P<0.0001);且FAP与MMP-1的表达也呈正相关(r为0.828;P<0.0001)。结论 FAP和MMP-1可能协同作用参与降解细胞外基质和促进肝星形细胞的活化,从而在肝纤维化的发生发展中起作用。
Objective To investigate the expression of fibroblast activation protein(FAP) and matrix metalloproteinase-1(MMP-1) in mouse hepatic fibrosis and the relationship between FAP and MMP-1,and the mechanism of hepatic fibrosis involved in them.Methods Experimental hepatic fibrosis model was induced by carbon tetrachloride.The degree of hepatic fibrosis was evaluated by HE and Masson's trichrome staining,and the dynamic expression of FAP and MMP-1 were detected by immunohistochemical technique in mouse hepatic fibrosis tissue.Results The expression of FAP and MMP-1 had the same location and extent in model group,especially in the fibroblasts of portal area and fibrous septa in mouse hepatic fibrosis tissue.The immunoreactivities of FAP and MMP-1 were positively correlated with the degree of hepatic fibrosis(r=0.672,0.684,P0.0001).Moreover,the expression of FAP had a positive correlation with MMP-1 in hepatic fibrosis tissue(r=0.828,P0.0001).Conclusion FAP and MMP-1 may play synergistic roles in the degradation of extracellular matrix(ECM) and the activation of hepatic stellate cells(HSCs),and play important roles in the development of hepatic fibrosis.
