摘要
目的探讨HtrA2(也称作0mi)在大鼠脑出血急性期的作用及其机制。方法雌雄各半SD大鼠150只,随机分为正常组、假手术组及脑出血后3h、6h、12h、1d、3d、5d组和7d组,每组10只。在脑立体定向仪下采用自体血注入尾状核法制备脑出血模型。于相应时间点处死动物,用干湿重法测量脑含水量(BWC);进行ICH组神经功能缺失评分;RT-PCR法检测HtrA2 mRNA表达水平。结果各组均有HtrA2 mRNA表达,但脑出血后各组手术侧表达明显增加(P<0.05),尤其是3d组,与脑含水量呈正相关;而脑含水量与神经功能缺失评分呈负相关。结论大鼠脑出血后HtrA2 mRNA表达增加,并与脑水肿程度呈正相关,推测HtrA2参与ICH后灶周围组织神经元凋亡的过程及ICH后灶周脑组织水肿的过程,诱导或加重了神经元的凋亡及脑水肿。
Objective To investigate the mechanism of HtrA2 in rat brain hemorrhage.Methods Evenly divided male and female SD rats 150,were randomly divided into control group,sham operation group and 3h after cerebral hemorrhage group,6h,12h,1d 3d,5d and 7d groups,n=10.In the stereotactic frame used under autologous arterial blood into the caudate nucleus preparation of intracerebral hemorrhage model.After animals were killed,ICH group for neurological deficit scores;By dry weight measurement of brain water content(BWC);RT-PCR assay expression of HtrA2 mRNA.Results The expression of HtrA2 mRNA in each group,but after intracerebral hemorrhage in each group operated side was significantly increased(P0.05),especially the 3d group,a positive correlation with brain water content;and brain edema and neurological deficit scores were negatively correlated.Conclusion Cerebral hemorrhage HtrA2 mRNA expression and the degree of brain edema was a positive correlation,Speculated that HtrA2 participate in ICH lesions surrounding tissues after the process of neuronal apoptosis and ICH perifocal brain edema after the process,induced or exacerbated neuronal apoptosis and cerebral edema.
