摘要
为开展水稻新品种亲缘关系鉴定,拓展水稻种质资源创新与利用途径,以筛选出的12条IS-SR引物对23个水稻品种(系)进行PCR扩增。结果表明,共扩增出清晰、重复性好的谱带155个,其中,特异带121条,多态性比例为78.06%;应用引物881、888及其组合获得了每个品种的ISSR特征带,可以有效区分出所有供试材料,建立了23份水稻品种(系)的ISSR指纹图谱;经聚类分析,在相似性系数为0.678处,可将所有材料严格地按粳、籼稻分为2类。
Twelve selected ISSR primers were used in PCR amplification of 23 tested rice cultivars to identify genetic relationship of new varieties,and expand innovation and utilization of germ plasm resource of rice.The results showed that 155 bands with distinct and good repeatability were amplified,121 specific bands were obtained and the polymorphic rate was up to 78.06%.The specific bands of each rice cultivar were obtained by applying primer 881,primer 888 and their combinations,which can effectively distinguish 23 tested rice cultivars to establish their ISSR finger-print.The clustering analysis showed that 23 tested rice cultivars could be strictly divided into two types of Indica and Japonica when the similar coefficient was 0.678.
