摘要
保健酒去乙醇处理后用盐酸溶液将其中的蛋白质和肽类化合物水解成游离态氨基酸,以避免酸水解过程中产生的不溶性气体导致水解管爆裂问题。采用柱前衍生反相高效液相色谱法测定游离氨基酸含量。在10~250μmol.L-1浓度范围内,16种氨基酸的峰面积和浓度之间的线性相关系数为0.982~0.999,16种氨基酸的定量下限(S/N=10)为0.5 mg.L-1,加标回收率为94%~101%,除蛋氨酸外,其它氨基酸的RSD均小于3.0%。
After removing ethanol,the protein and peptide in healthcare wine were decomposed into free amino acids by hydrolysis with hydrochloric acid.Removing ethanol could avoid pipe bursting because of the formed insoluble gas during hydrolysis processing.The content of free amino acids were determined by pre-column derivatization and reversed-phase high performance liquid chromatography(RP/HPLC).The calibration curves of 16 amino acids were linear in the range of 10-250 μmol·L-1 with correlation coefficients of 0.982-0.999.The limits of quantitation were 0.5 mg·L-1.The spiked recoveries of 16 amino acids were in the range of 94%-101%,and the RSDs for all amino acids were less than 3.0% except for methionine.
