摘要
目的:初步探讨一次性跑台运动影响肥胖大鼠摄食的神经内分泌机制。方法:10周成功建模营养性肥胖SD大鼠46只,分为对照组(C)和运动后即刻组(E0)、1小时组(E1)、3小时组(E3)、12小时组(E12)和24小时组(E24)。各运动组大鼠在专用电动跑台上进行一次性运动,跑台坡度5%,速度20m/min,时间为40min。分别于运动后即刻、1h、3h、12h和24h观察大鼠摄食量、血糖、血脂指标;采用放射免疫法测试血浆和下丘脑ghrelin和obestatin蛋白表达,RT-PCR方法测试下丘脑ghrelin受体GHSR-1α、obestatin假定受体GPR-39的基因表达。结果:一次性跑台运动后,与对照组相比,E1、E3、E12和E24组大鼠摄食量下降,血糖和血脂也有不同程度的下降趋势,其中E1组血糖和E12组血脂下降显著;与对照组比较,运动后不同时间段大鼠血浆ghrelin和obestatin无明显变化,而运动后即刻下丘脑ghrelin和obestatin显著下降(P<0.01),在12h和24h又明显上升(P<0.01);与对照组相比,运动后各组下丘脑GHSR-1α基因表达无明显差异,且未见下丘脑GPR-39基因表达。结论:Ghrelin影响大鼠摄食和能量代谢调节可能是通过下丘脑中枢机制,而与外周血ghrelin无关。
Objective The aim of this paper is to study the effects of acute exercise on the appetite-related hormones levels in plasma and hypothalamus of obese rats. Methods Diet-induced obese rats were randomly divided into six groups,group C(control),group E0(immediate after exercise),group E1(1 hour after exercise),group E3(3 hours after exercise),group E12 (12 hours after exercise) and group E24 (24 hours after exercise). The rats in exercise groups ran on treadmill at a speed of 20m·min-1 with a slope of 5% for 40 minutes. The meal size and blood glucose (Glu)and triglyceride(TG)of all rats were studied. Concentrations of ghrelin and obestatin were measured by RIA. Expression of ghrelin receptor(GHSR-1α)and putative obestatin receptor(GPR-39)in the hypothalamus were measured by quantitative RT-PCR. Results After acute exercise,comparing to group C,meal size decreased in groups E1 and E3,and normal in groups E12 and E24. Concentrations of Glu and TG exhibited a downward tendency after exercises (Glu decreased sharply in groups E1 and E12). Ghrelin and obestatin in plasma remained unchanged,whereas decreased in hypothalamus in groups E0,E1 and E3,and increased in groups E12 and E24 (P 0.01). These changes were consistent with the meal size of obese rats. After acute exercises,GHSR-1α remained unchanged and GPR-39 was not found in hypothalamus. Conclusion The appetite of obese rats decreases after acute exercises because of reduced level of ghrelin in the hypothalamus.
