乳腺导管癌淋巴转移拟态组织芯片的构建

摘要 目的建立一套模拟乳腺导管癌淋巴结转移过程的组织芯片。方法选用16例微小浸润性导管癌标本中的癌旁组织、导管原位癌和微小浸润癌组织;32例无淋巴结转移的浸润性导管癌标本中的非浸润性癌、浸润性癌和淋巴管癌栓以及32例淋巴结转移性浸润性导管癌标本中的非浸润性癌、浸润性癌、淋巴管癌栓和淋巴结转移性癌组织。按肿瘤发生发展的顺序制成组织芯片。常规石蜡切片,HE染色及免疫组织化学染色。结果组织芯片所包含的信息和供体蜡块一致,组织芯片免疫组织化学染色结果与普通切片一致。结论成功构建了乳腺导管癌淋巴结转移拟态组织芯片,为乳腺癌淋巴结转移机制的深入研究提供了良好的平台。 Objective To establish a serial of tissue chips to simulate lymph node metastases in breast duct carcinoma. Methods Perieancerous tissues, duetal carcinoma in situ (DCIS) and micro - infiltrating carcinoma from 16 cases of microinvasive ductal carcinoma(MIDC) specimens, non - invasive ductal carcinoma, invasive ductal carcinoma and lymphatic vessel tumor emboli from 32 cases of non lymph node metastasis invasive ductal carcinoma specimens, and non - invasive ductal carcinoma, invasive ductal carcinoma, lymphatic vessel tumor emboli and lymph node metastasis carcinoma from 32 cases of lymph node metastasis invasive ductal carcinoma specimens were selected and made into tissue chips in order of the sequences of the tumorigenesis and development of the tumor. Then, paraffin sections, HE staining and immnohistochemical staining proceeded. Results The information of tissue chip coincided with that of donator paraffin block on slice. Immnohistochemical staining result of tissue chip also coincided with that of donator paraffin block. Conclusion Mimetic tissue chips of lymph node metastases in breast duct carcinoma were established successfully, which offered an intensive research platform for lymph node metastasis mechanism in breast duct carcinoma.
